Two buses: the local bus from Kalampaka to Trikala, then the KTEL Trikalon (KTEL Trikala) coach from Trikala to Thessaloniki's Macedonia terminal, about 2 h 30. Allow about three and a half hours in all. Both legs are below with their live timetables.
Coaches leave Trikala for Thessaloniki through the day, the last at 20:00, and the run to the Macedonia terminal takes about 2 h 30. After a morning at the monasteries, the local bus from Kalampaka to Trikala and an afternoon coach gets you into Thessaloniki by early evening. Allow at least half an hour at Trikala between the two buses.
The Macedonia terminal is west of the centre, with city buses and taxis outside. If you are continuing to the Chalkidiki beaches, note that those buses leave from a different station, KTEL Chalkidikis at the 9th km, not from the Macedonia terminal.
